Mercaptopurine is an inactive pro-drug which acts as a purine antagonist but requires cellular uptake and intracellular anabolism to thioguanine nucleotides for cytotoxicity. The mercaptopurine metabolites inhibit de novo purine synthesis and purine nucleotide interconversions.
