Indication for Naldemedine

Population group: only adults (18 years old or older)

Naldemedine is indicated for the treatment of opioid-induced constipation (OIC) in adult patients who have previously been treated with a laxative.

Detailed description

The recommended dose of naldemedine is 200 microgramm daily. Naldemedine may be used with or without laxative(s). It may be taken at any time of the day but it is recommended to be taken at the same time every day.

Alteration of the analgesic dosing regimen prior to initiating naldemedine is not required. Naldemedine must be discontinued if treatment with the opioid pain medicinal product is discontinued.

Dosage considerations

Naldemedine should be taken once daily, with or without food.

Naldemedine is an antagonist of opioid binding at the mu-, delta-, and kappa-opioid receptors. Naldemedine functions as a peripherally-acting mu-opioid receptor antagonist in tissues such as the gastrointestinal tract, thereby decreasing the constipating effects of opioids without reversing the central nervous system (CNS)-mediated opioid effects.
