Indication for Rilonacept

Population group: only adolescents (12 years - 18 years old)

Rilonacept is indicated for the treatment of Cryopyrin-Associated Periodic Syndromes (CAPS) with severe symptoms, including Familial Cold Autoinflammatory Syndrome (FCAS) and Muckle-Wells Syndrome (MWS), in adults and children aged 12 years and older.

2.2-4.4 mg/kg once weekly

Detailed description

Treatment should be initiated with a loading dose of 4.4 mg/kg, up to a maximum of 320 mg. Dosing should be continued with a once-weekly injection of 2.2 mg/kg, up to a maximum of 160 mg. Dosing in children must be adjusted as the child grows. The patient or care giver should be advised to speak to the treating physician before adjusting the dose. The experience in children is limited. In the clinical program for CAPS, 8 adolescents aged 12-17 were treated for up to 18 months.

Rilonacept is a dimeric fusion protein which binds to and blocks the activity of the cytokine IL-1 and binds both IL-1β and IL-1α. Rilonacept is indicated for the treatment of Cryopyrin-Associated Periodic Syndromes (CAPS) with severe symptoms.
